IV: Clinical Trials of GP IIb/IIIa Inhibition

  • The PURSUIT (Platelet glycoprotein IIb/IIIa in Unstable Angina: Receptor Suppression Using Integrilin Therapy) study was a double-blind randomized controlled trial in 10,948 patients with NSTEMI to determine whether eptifibatide added to antithrombotic therapy with heparin and aspirin would provide an incremental clinical benefit.

  • Patients were randomly assigned to treatment with eptifibatide at a high or low dose, or placebo. High-dose eptifibatide was administered with a 180 µg/kg bolus, then 2.0 µg/kg per min for 72 hours. A lower-dose eptifibatide treatment group was stopped after safety of the high dose was shown in accordance with the protocol.

  • The primary outcome was the effect of high-dose eptifibatide vs placebo on the composite of death and nonfatal MI up to 30 days.
